Salisbury station is one of the busiest stations in the Adelaide metropolitan area. Most trains stop at this station. Some trains also run non-stop to Adelaide from Salisbury and in the opposite direction to Gawler Central. It forms part of an interchange where passengers can also change for buses.

Salisbury station was the site of an accident that occurred in October 2002. Four people were killed when a train bound for Alice Springs (The Ghan) collided with a bus and a car on the Park Terrace level crossing adjacent to Salisbury station.
